How to Reconstitute Peptides: Complete Step-by-Step Guide
A physician-approved guide to safely mixing lyophilized peptide powder with bacteriostatic water, drawing your dose, and performing a subcutaneous injection.
What You'll Need
Lyophilized peptide vial
Your prescribed peptide in powder form
Bacteriostatic water
Sterile water with 0.9% benzyl alcohol preservative
10 mL mixing syringe
Large syringe with needle for reconstitution
Insulin syringe (30-31 gauge)
Fine needle for subcutaneous injection
Alcohol wipes
Sterile prep pads for cleaning vial tops and skin
Sharps container
For safe disposal of used needles
Part 1: Reconstitution (Mixing)
Reconstitution is the process of dissolving your lyophilized (freeze-dried) peptide powder into bacteriostatic water to create an injectable solution. This must be done carefully to preserve the peptide's integrity.
Step 1: Prepare Your Workspace
Work on a clean, flat surface. Wash your hands thoroughly. Flip the protective caps off both the bacteriostatic water vial and the lyophilized peptide vial.
Step 2: Sterilize the Vial Tops
Wipe the rubber stopper of each vial with a fresh alcohol wipe. Use a new wipe for each vial. Allow to air dry for a few seconds.
Step 3: Draw Bacteriostatic Water
Take the large 10 mL syringe with needle out of its wrapping. Make sure the needle is securely attached by pressing and twisting. Pull back the plunger to the volume your healthcare provider has prescribed (this determines your concentration).
How much water should you add?
The amount of bacteriostatic water determines your concentration. More water = more dilute solution = easier to measure small doses. A common ratio is 2 mL of water per 5 mg vial. Use our Reconstitution Calculator to determine the exact amount for your prescribed dose.
Step 4: Transfer Water to Peptide Vial
Insert the needle through the rubber stopper of the bacteriostatic water vial. Flip the vial upside down and push the plunger to transfer the air into the vial. Then pull back the plunger to draw your prescribed amount of water into the syringe.
Step 5: Add Water to the Peptide
Insert the syringe into the rubber stopper of the lyophilized peptide vial. Hold at a slight angle and slowly press down on the plunger to inject the bacteriostatic water. Let the water run down the inside wall of the vial — never spray directly onto the powder, as this can damage the peptide.
Step 6: Dissolve
Once all the water has been added, carefully remove the needle and discard it in a sharps container. Gently swirl the vial until all powder has dissolved into the solution. Never shake — shaking can denature the peptide and reduce its effectiveness.
Important: Never Shake Your Peptides
Peptides are fragile protein chains. Vigorous shaking can break molecular bonds and destroy the active compound. Always gently swirl or roll the vial between your palms.
Part 2: Drawing Your Dose & Injection
Step 7: Sterilize Again
Wipe the top of the reconstituted peptide vial with a fresh alcohol wipe. Then wipe your injection site with another new alcohol wipe. Common injection sites include the lower abdomen (around the belly button) and the thigh.
Step 8: Draw Your Dose
Take your 30 or 31 gauge insulin syringe and carefully remove both caps. Pull back the plunger to the mark prescribed by your healthcare provider. Insert the needle through the rubber stopper of the peptide vial.
Flip the vial upside down and push the plunger to transfer air into the vial. Then pull back to draw the prescribed amount of peptide solution into the syringe. Check for air bubbles — if present, gently tap the syringe and push them back out.
Step 9: Inject
Pinch the injection area on your body about 2 inches high and wide to create a fold of skin and subcutaneous fat. Hold the needle at a 90-degree angle, pierce the skin, and carefully inject the medication by slowly pressing the plunger. Then carefully remove the needle and dispose of it in a sharps container.
Storage Guidelines
| State | Storage | Duration |
|---|---|---|
| Unreconstituted (powder) | Refrigerator or freezer | Months to years |
| Reconstituted (mixed) | Refrigerator (36-46°F / 2-8°C) | Up to 4 weeks |
| During travel | Insulated cooler with ice packs | Keep cold, use within hours |
Common Mistakes to Avoid
- Shaking the vial — Always gently swirl. Shaking denatures peptides.
- Spraying water directly on powder — Aim down the vial wall at an angle.
- Reusing needles — Always use a fresh needle for each injection. Reuse increases infection risk and dulls the needle.
- Skipping the alcohol wipe — Sterilizing vial tops and injection sites prevents contamination.
- Storing at room temperature — Reconstituted peptides must stay refrigerated.
- Guessing your dose — Use the reconstitution calculator or follow your physician's exact instructions.
